Cell Counting

typanblue, hemocytometer, miro tube, pipette

 

Method

1. trypanblue 2  : cell 1  비율 로 miro tube에 넣고 pipetting.

2. 20㎖를 hemocytometer에 넣고 cover glass로 넣고 cover glass로 덮은 후 cell counting.

 

계산 : counting한 cell 개수 × 3 × 1/4 × 10⁴ 

빨간 선에 닿은 cell은 counting해주고, 검정 선에 닿은 cell은 counting하지 않는다.

 

계산법

3 : trypanblue와 cell의 비율은 2:1로 하였기에 이를 희석 시켜주어야 하기에 3을 곱해주는 것이다.

1/4 : counting한 구역이 4개 이기에 평균을 만들어 주기 위해

10⁴  : cell counting 단위가 ㎖이고, hemocytometer의 한 칸당 100nl 즉  1㎖당 10⁴nl이기에

죽은 세포를 파랗게 염색 시켜 counting할 때 염색된 세포는 counting하지 않는다.
